【目的】褐飞虱是危害水稻的主要虫害之一，利用新的抗性基因和通过基因聚合培育抗性品种是目前防治褐飞虱最经济有效的方法之一。【方法】由药用野生稻衍生来的水稻品系B5 对褐飞虱生物型1 和2 具有高度抗性，B5 携带的抗性基因Bph14 和Bph15 分别被定位在第3 和第4 染色体上，本研究以B5 为抗褐飞虱供体亲本，以优良杂交稻亲本9311 和1826 为受体亲本，通过复交和回交，分别利用与Bph14 和Bph15 紧密连锁的SSR 标记 MRG2329 和MS5 在分离群体中进行分子标记辅助选择，通过苗期分子标记检测和成株期农艺性状选择，【结果】最终获得一系列目标基因纯合且农艺性状优良的稳定株系。采用苗期群体鉴定技术，对其中的38 份重点株系进行了抗虫鉴定，在26 份Bph14 单基因纯合株系中，除2 份为中感外，92.31%的材料抗性水平在中抗以上，6 份Bph15 单基因纯合株系全为抗或高抗，聚合有Bph14 和Bph15 双基因的6 份株系抗性都达到了高抗水平，抗性强于Bph14 或Bph15 单基因纯合株系。【结论】研究表明在育种进程中利用与Bph14 和Bph15 紧密连锁的分子标记开展抗褐飞虱分子标记辅助育种是一种有效的途径。
【Objective】 The brown planthopper (BPH) is an important insect pest of rice. One crucial measure for BPH control is to breed resistance cultivars through identification of new BPH resistance genes and pyramiding major resistance genes.【Method】 B5 is a rice line derived from the wild rice Oryza officinalis and is highly resistant to the BPH biotypes 1 and 2. The dominant BPH resistance genes Bph14 and Bph15 have been identified in B5 and mapped on rice chromosome 3 and chromosome 4, respectively. In this study, we used B5 as a BPH resistance donor. The elite restorer lines 9311 and 1826 were used as recipients of the BPH resistance genes in B5. Screened SSR markers MRG2329 and MS5 were used to select BPH resistance genes in the segregating populations. 【Result】 We performed molecular marker-assisted selection (MAS) at the seedling stage and agronomic trait selection at the adult plant stage. We obtained some introgression lines that carried BPH resistance genes and stable agronomic traits. The BPH resistance levels in 38 important lines were determined in the mass seedling screening test. Among 26 Bph14-single introgression lines, 23 lines were resistant or highly resistant, 1 line was moderately resistant, and 2 lines were moderately susceptible. All of the 6 Bph15-single introgression lines showed resistance or high resistance. However, all the 6 Bph14/Bph15 pyramided lines were highly resistant to BPH.【Conclusion】The results indicated that 92.31% Bph14-single introgression lines had moderate resistance to the BPH insect, and the Bph14/Bph15 pyramided lines had higher resistance levels than the single-gene introgression lines. This survey also revealed that the two markers linked with Bph14 and Bph15 were effectively utilized in BPH-resistant breeding programs.
