为了阐明高抗条纹叶枯病水稻品种抗病性的分子机理,以江苏省高抗条纹叶枯病粳稻品种‘镇稻88’和‘徐稻3号’为研究材料,应用双向电泳联用质谱技术对2种水稻苗期蛋白质表达谱的特点及差异进行比较,并对差异蛋白进行了鉴定以及分析归类。结果表明:2个水稻品种的蛋白图谱上均检测到大约1000个蛋白点,这些蛋白点主要分布在等电点为4～7,分子量为10～100kDa的范围内。用ImageMaster 2D Platinum软件分析后发现,2个水稻品种间共存在7个差异蛋白点,在‘徐稻3号’中表现为下调表达。质谱分析共鉴定出6种蛋白,这些蛋白包括能量相关蛋白、代谢相关蛋白、防御相关蛋白和未知功能蛋白。
Rice stripe disease is one of the most economically devastating and widespread viral disease of rice(Oryza sativa L.). To illustrate the molecular mechanism of high resistance rice varieties to rice stripe diseaseresistance, the two-dimensional electrophoresis and mass spectrometry were used to study the characteristicsand differences of protein expression profiles between rice cultivars of‘Zhendao88’and‘Xudao3’, which hadhigh resistance to rice stripe disease. The results showed that approximately 1000 protein spots were separatedand visualized on coomassie brilliant blue (CBB) stained two-dimensional electrophoresis (2-DE) gels, whichwere mainly distributed in the isoelectric point of 4 to 7, 10-100 kDa molecular weight range. With the help ofImageMaster 2D Platinum software, 7 down-regulated proteins were found in‘Xudao3’and 6 proteins wereultimately identified by MALDI-TOF. These proteins included energy associated proteins, metabolismassociated proteins, defense associated proteins, and unknown protein.
