【突变体表型】

水稻突变体d61对外施BR不敏感,体内BR信号传导受阻,其典型表型有:特定节间的伸长受限;叶角减小,叶片直立,叶鞘比野生型短;穗颈比野生型长;黑暗中的形态建成受阻。d61的表型与OsBRI1基因的突变有关。

【定位与克隆】

利用已克隆的编码拟南芥BR受体激酶的BRI1基因,在水稻中发现了同源的OsBRI1,连锁分析表明OsBRI1d61等位,位于水稻第1染色体长臂,与RFLP标记C1370紧密连锁(Yamamuro et al. 2000)。

OsBRI1 cDNA全长3783 bp,仅有1个外显子,编码一个由1211氨基酸组成的BR 受体激酶。其中,d61-1是由于编码989位氨基酸的C→T导致苏氨酸变为异亮氨酸,而苏氨酸位于OsBRI1激酶的第6保守子域;d61-2则是由于编码第491位氨基酸的G→A导致缬氨酸替换为甲硫氨酸(Yamamuro et al. 2000)。

【生物学功能】

OsBRI1是一个BR信号受体,与拟南芥BRI1具有很高的序列相似性,在水稻的生长和发育中起多种作用:1. 节间伸长,包括诱发居间分生组织的形成和节间细胞的纵向伸长;2. 叶倾角,即弯曲度;3. 暗形态建成(Yamamuro et al. 2000)。

OsBRI1通过控制细胞分裂和伸长,调控器官发育,但对器官起始不是必需的(Nakamura et al. 2006)。
